The invention relates to purified human DNase having the amino acid sequence depicted in fig. 1, where the Asn residue corresponding to Asn-74 position from the human DNase is deamidated or non-deamidated, as well as to variations thereof, resulting from punctiform mutations, and being enzymatically active in DNA hydrolysing, and to a process for preparing purified human DNase deamidated at the residue of Asn-74 from a mixture of deamidated and non-deamidated human DNase, by separation with tentacle cation exchange resin, or with mobilised heparin resin or with an analogous non-hydrolysable mobilised DNA resin, and to a composition of purified DNase.
