A method of preparing a mutated gene is provided, comprising transforming a host cell with a phagemid containing a gene of interest; subjecting the host cell to a mutational event to produce a mutation in the DNA of the phagemid; infecting the host cell with a suitable helper phage to initiate packaging of the DNA; and infecting a second host cell with the resultant mixture of phage particles.
