摘要 |
To provide the followings that are applicable for in vitro cell-differentiation systems other than ES cells: cells whose function of expressing normal genes (wild type genes) is deficient or suppressed in the alleles corresponding to trapped genes, and the preparation method for the cells; a gene-trap method using the cells; and antisense promoter insertion vector for use in preparation of the cells. Not only trap-clone is efficiently obtained by introducing a gene-trap vector into the cells, but antisense RNA to the trapped genes are forcibly transcribed and the transcipts from wild type genes are disrupted by introducing, in inverse direction to the endogenous gene, antisense promoter insertion vector which is capable of inserting a strong promoter in a certain position of the mutant locus where the gene-trap vector of the trap-clone is inserted in. <IMAGE> |
