PROBLEM TO BE SOLVED: To obtain a protein having Junctophilin activity needed for constructing a junction membrane complex in a muscle cell of skeletal muscle, or the like, or a cell of neuron, or the like; a gene encoding the protein; a method for diagnosing diseases associated with the expression or activation of a protein having Junctophilin activity by using the gene; and the like. SOLUTION: Hybridomas are prepared by using membrane vesicles of rabbit skeletal muscle according to a routine procedure, the immunoblot analysis of microsome protein is carried out by using a monoclonal antibody obtained from the hybridomas, and thereby a protein of rabbit JP-1, that is, a triplicate zygote, is isolated and identified. Further, the protein of rabbit JP-1 consists of large membrane regions which link junction SR membranes and hydrophobic segments of C-terminal, and the cytoplasm region (N-terminal side) can bind to a cell surface membrane and exhibits affinity selectively to a phospholipid of the membrane component. |
