摘要 PROBLEM TO BE SOLVED: To detect the mutation in the base sequence of the nucleic acid in high throughput in no need of an expensive apparatus. SOLUTION: A solution including a double-stranded nucleic acid is heated to dissociate into the single strand, then cooled down to allow them to conjugate into the double strands. In the case where the double-stranded nucleic acid is the homo type, the homo type double-stranded chain and the hetero type double stranded chain are formed, in the case where the doublestranded nucleic acid is the homo type, only the homo type double stranded chain is formed. As the temperature of the solution is raised, the heat-fusing curve is depicted by measuring the ultraviolet absorption. In the case where the double stranded chain is only the homo double-stranded chain after the heating and the cooling, the curve has one or close tow Tm temperature, while in the case where the double-stranded chain includes both of the homo and hetero type double-stranded chains, the curve has apart two Tm temperatures and further one Tm temperature individually near the parted Tm temperatures, respectively. The resultant heat-fusing curve is primarily differentiated with absorbance to know the inflection point, in other words, the Tm temperature. The existence of the hetero double stranded chains are detected with Tm temperature itself or the number of the Tm temperatures, in its turn, the mutation of the base sequence of the double- stranded chin nucleic acid is detected.
