Retroviral packaging plasmid for production of recombinant retrovirus in humans

A method of transducing mammalian haematopoietic stem cells with retroviral supernatants produced by transient transfection is disclosed. The method involves a transient cotransfection of a first population of mammalian cells that can produce a virus with a retroviral helper DNA sequence and a retroviral vector encoding a foreign gene. This enables the population of mammalian cells to produce replication-defective recombinant retroviral vectors carrying the foreign gene that can found in the cell supernatant. A second population of mammalian cells is grown on culture plates with adhesion molecules or antibodies to adhesion molecules, and is incubated with the supernatant collected from the first population of mammalian cells. The cells transduced with the foreign gene are also claimed.
