Shuffling of heterologous DNA sequences using conserved regions

Disclosed is a method of shuffling heterologous DNA sequences. It has use in, for example, creating enzymes with a desired function from similar enzymes that collectively have these functions. The method involves identifying conserved regions of the heterologous DNA sequences, generating primers to those regions, creating fragments from the heterologous DNA using those primers, and shuffling the generated fragments.
