An electrophoresis system for separating small macromolecules comprising two electrophoretic systems, wherein each system comprises an anode buffer and cathode buffer chamber containing an anode and cathode therein; an ion-permeable separation membrane positioned between the anode and cathode buffer chambers; an ion-permeable restriction membrane positioned either side of the ion-permeable separation membrane to define first and second interstitial volumes; an electric field applied between the buffer chambers, and wherein the two electrophoretic systems are in fluid communication with each other.
