An oil-, polar lipid-, and protein-containing mixture is fractionated by adding a water-soluble organic solvent to the mixture and separating protein from the mixture to form a protein-rich fraction and a polar lipid/oil-rich fraction, and subjecting water/water-soluble organic solvent and polar lipid/oil-rich fraction to density separation. Fractionation of an oil-, polar lipid-, and protein-containing mixture involves adding a water-soluble organic solvent to the mixture and separating protein from the mixture to form a protein-rich fraction and a polar lipid/oil-rich fraction. The concentration of water-soluble organic solvent in the polar lipid/oil-rich fraction is reduced. The water/water-soluble organic solvent and polar lipid/oil-rich fraction is subjected to density separation to form a polar lipid-rich fraction and an oil-rich fraction.
