PROBLEM TO BE SOLVED: To provide a method for rapidly assaying a protease with a high accuracy in which the adaptation can be made even to inspection, etc., of a biosample during operations. SOLUTION: This method is to assay the protease activity and comprises (1) a step of bringing a sample containing the protease into contact with a thin film formed on the surface of a support, containing at least one kind of protease substrate and cross-linked and/or substantially insoluble in water and (2) a step of washing the thin film with water and detecting a digestion mark formed in the film by actions of the protease in which the thin film contains 0.001-1 mol/m2 calcium ions.
