发明名称 Nouveau sel de l'acide glucose-1-phosphorique et procédé de préparation
摘要 1,054,493. Salt of glucose-1-phosphoric acid. ROUSSEL-UCLAF. June 30, 1964 [July 9, 1963], No. 27010/64. Heading C2C. The invention comprises calcium glucose-1- phosphate of the formula and its dihydrate. They may be prepared by reacting a compound containing glucose-1- phosphate ions with a compound containing calcium ions preferably in an aqueous medium. A basic calcium compound e.g. calcium hydroxide which may be in the form of milk of lime may suitably be reacted with glucose 1- phosphoric acid. An alternative procedure is to react another salt of glucose-1-phosphoric acid with a water-soluble calcium salt. The salt of glucose-1-phosphoric acid may be the potassium, sodium, barium or ammonium salt, and the calcium salt is conveniently the chloride. A further process comprises transforming a salt of glucose-1-phosphoric-acid into the free acid by reaction with a cation exchange resin in the acid form, and then neutralizing the free acid with a basic calcium salt, or the salt of glucose- 1-phosphoric acid may be adsorbed on to an anion exchange resin in the salt form, converted into ammonium glucose-1-phosphate by elution with ammonia and then reacted with a soluble calcium salt. The calcium glucose-1-phosphate from any of these processes may be isolated by precipitation from aqueous solution with ethanol or another water-miscible organic solvent, by lyophilisation or by crystallization from a concentrated aqueous solution. Calcium glucose-1-phosphate may be used as the active ingredient in pharmaceutical compositions. The compositions may be in solid or liquid form. Preferably the solid forms are in dosage units containing from 250 mg. to 1 g. of active ingredient e.g. in the form of tablets, pills, dragees and cachets, and suppositories for rectal administration. Suitable carriers or excipients for solid compositions include lactose, starches, silica and magnesium stearate and suppository bases e.g. cocoa butter or a glyceride. Liquid compositions may be adapted for oral or parenteral administration. The liquid compositions for oral administration include syrups, suspensions, elixirs and emulsions. Liquid compositions for parenteral administration require a parenterally acceptable liquid or oil as carrier e.g. sterile, pyrogen-free water or arachis oil. Calcium glucose-1-phosphate may be used in the treatment of decalcification, rickets, dental carriers, fractures, osteoporosis, tuberculosis, states of debility, loss of memory, anaemia and during convalescence from infectious diseases.
