PROBLEM TO BE SOLVED: To provide a kit for detecting the destruction rate of erythrocyte in a sample by the utilization of the peroxidase activity of hemoglobin, and to provide a method for correcting the affect of an assay target substance to the assay values by the destruction of the erythrocyte, such as hemolysis or occult blood, and for accurately assaying the assay target substance except the erythrocyte in the sample. SOLUTION: This kit for the assay, comprising (1) a peroxidase (standard), (2) hydrogen peroxide, and (3) a coloring agent. The method for assaying the assay target substance except erythrocyte in a sample, characterized by measuring the activity of a peroxidase in the sample to detect the destruction rate of the erythrocyte, determining the concentration of the assay target substance originated from the erythrocyte by the destruction of the erythrocyte from the obtained destruction rate of the erythrocyte and the concentration of the assay target substance per the volume of the erythrocyte, and then subtracting the concentration of the assay target substance originating from the erythrocyte from the assayed concentration of the assay target substance.
