Preparation of perbenzylated 1-O-glycosides comprises reacting a perbenzylated 1-OH-sugar with a protected alkylating reagent in an organic solvent at 0-50 deg C in the presence of a base and optionally also a phase-transfer catalyst, followed by splitting off the protective group and optionally converting the product to a salt. Preparation of perbenzylated 1-O-glycosides of formula (I) comprises reacting a perbenzylated 1-OH-sugar of formula (II) with a protected alkylating reagent of formula (III) in an organic solvent at 0-50 deg C in the presence of a base and optionally also a phase-transfer catalyst, followed by splitting off the protective group and optionally converting the product to a salt. Sugar<1> = a monosaccharide functionalized in the 1-position; R = benzyl; n = 2, 3 or 4; X = -O-, -S-, -COO- or -NH-; and L = 1-30C alkyl optionally unsaturated and optionally interrupted by 1-10 O or 1-3 S atoms, 1-2 phenylene, phenyleneoxy or phenylenedioxy groups or a thiophene, pyrimidine or pyridine residue and/or substituted by 1-3 phenyl, carboxyl, amino or CF3 groups, 1-5 OH or O-(1-7C)alkyl groups or 1-10 F atoms; Nu = a 'nucleofug' such as -Cl, -OTs or - OSO2CF3; and Sg = a protective group.
