<p>Disclosed are high throughput assays for rapidly screening a library of nucleic acid molecules to identify a gene product that modulates expression of a gene of interest. The assays generally involve (a) introducing into one or more plant protoplasts (i) a reporter gene construct operably linked to a promoter of a gene of interest and (ii) a member of a library of nucleic acid molecules, wherein the library member is expressed in the plant protoplasts; and (b) screening the protoplasts to determine whether the amount of gene expression of the reporter gene construct changes in response to the expression of the library member, a change in gene expression of the reporter gene construct identifying the gene product expressed by the library member as one that modulates expression of the gene of interest.</p> |
