PROBLEM TO BE SOLVED: To provide a method and instrument for more accurately measuring the concentration of a substance contained in a specimen, which contains many components in a mixed state, in a very small amount. SOLUTION: At first, the specimen containing molecules to be measured is intermittently ejected into a vacuum chamber 12 reduced in pressure to form a molecular beam jet 13j and the vibrating rotation-temperature of the specimen is cooled to about several 10K or lower to make the absorption spectrum wavelength width of the molecules to be measured sufficiently separable from the absorption wavelength of coexisting molecules. Next, the molecular beam jet 13j is irradiated with exciting light having a wavelength coinciding with the absorption spectrum wavelength width of the molecules to be measured through an introducing optical fiber 15. Then, the intensity of fluorescence of the molecular beam jet 13j induced by the exciting light is measured to judge the concentration of molecules to be measured present in the molecular beam jet 13j.
