The present invention comprises method of detecting biomolecules <i>in situ</i> in gel separation medium following electrophoresis that provides sensitivity similar to blotting. This method is applicable to <i>in situ</i> detection of a wide range of biomolecules including proteins and nucleic acids. After electrophoretic separation of a sample in gel separation media comprising a gellable polymeric material other than cross-linked polyacrylamide, the gel is contacted with a solution comprising at least one detectably labeled reagent directed to a biomolecule under conditions suitable for binding of the reagent to the biomolecule. Alternatively, the gel is contacted with a solution comprising at least one non-detectably labeled reagent directed to a biomolecule and a solution comprising at least on detectably labeled reagent directed to a biomolecule. In either case, binding of the detectably labeled reagent is assessed and indicates detection of biomolecules in the gel.
