PROBLEM TO BE SOLVED: To obtain an oligonucleotide having largely excessive diastereomers and containing an optically isomerically pure phosphorothioate bond, a phosphorus-bonded oligonucleotide containing substantially whole Rp bonds or whole Sp bonds, a research material and a diagnostic material for examining a physical state of animal, especially a disease state and a sequence-specific oligonucleotide containing am optically isomerically pure phosphorothioate bond and to provide a method for synthesizing an intermediate useful for the synthesis of the oligonucleotide. SOLUTION: This method synthesizes a phosphorothioate bond oligonucleotide rich in diastereomers. This intermediate is useful for the synthesis. This method chemically synthesizes a sequence-specific phosphorothioate oligonucleotide containing a chiral phosphorus bond and other oligonucleotides.
