Provided is the use of scaffold attachment regions (SARs) to increase gene expression in primary no-proliferating cells or resting cells, particular in transduced eukaryotic resting primary cells, especially retrovirally transduced cells. The SAR sequence is reported to have no detectable influence on retroviral vector expression in transduced cell lines. Specifically claimed is a retroviral vector comprising genetic material corresponding to (a) at least one SAR, and (b) at least one heterologous gene operatively linked to an expression control sequence, the heterologous gene (or at least one if there are more than one heterologous gene) being rev-M10 and the SAR or at least one SAR is derived from, obtainable from or corresponds to the 5' SAR of the human interferon-b gene, as well as claiming cellular compositions comprising non-immortal human cells transduced with the retroviral vector, particularly hematopoietic cells like T cells. The retroviral vector can be used to prepare compositions for the treatment of HIV infection.
