Described is: a) A purified and isolated polypeptide, capable of binding to the extracellular domain of Flt4 receptor tyrosine kinase.(Fit4), the polypeptide being encoded by plasmid pFLT4-L, deposited as ATCC Accession Number 9723. b) A purified and isolated polypeptide, capable of binding to the extracellular domain of Flt4 receptor tyrosine kinase comprising an amino-acid sequence (a contiguous portion of SEQ ID NO: 33 in the specification) effective to permit binding, wherein the contiguous portion includes eight cysteine residues that are conserved in human vascular endothelial growth factor (VEGF), human platelet derived growth factor A (PDGF-A), and human platelet derived growth factor B (PDGF-B), and wherein the polypeptide lacks any portion of the SEQ ID NO: 33 that has one or more cysteine motifs of Balbiani ring 3 protein (BR3P). c) Purified proteins comprising two of the polypeptide ligands (from a or b above) linked together. d) Nucleic acids comprising a nucleotide sequence encoding a polypeptide as described in a) or b) above. e) Vectors comprising a nucleic acid as described in d), above. f) A host cell transformed or transfected with a nucleic acid as described in d), above. g) A method of making a polypeptide capable of binding to the extracellular domain of Flt4 receptor tyrosine kinase. h) An antibody that is specifically reactive with a polypeptide as described in a) or b) above. i) A polyclonal antiserum produced by immunizing a mammal with a composition comprising a polypeptide as described in a) or b) above, then obtaining the antiserum from the mammal. j) A method for detecting endothelial cells in a biological sample by exposing a biological tissue sample comprising endothelial cells to a polypeptide as described above under conditions where the polypeptide binds to endothelial cells; and detecting the polypeptide bound to endothelial cells in the biological tissue. k) An in vitro method of modulating the growth of mammalian endothelial cells by exposing mammalian endothelial cells in vitro to a polypeptide as described above. l) A purified and isolated polypeptide which is capable of binding the extracellular domain of Kdr receptor tyrosine kinase (VEGFR-2), the polypeptide having an amino acid sequence comprising a portion of SEQ ID NO: 33. The above described polypeptides ("a)", "b)", "l)"), proteins, nucleic acids, vectors and antibodies; are useful for modulating the growth of mammalian and/or lymphatic endothelial cells.
