摘要 A new method (M1) of analysis of differential gene expression comprises competitive hybridization of polynucleotide (I) populations (A) of expressed genes, from two different cell or tissue sources, with a reference population (B) of sequences attached to separate solid phase supports in clonal subpopulations. In M1, each (I) carries a light-generating label, different for each source, and duplexes between (A) and (B) are formed at ratios directly proportional to the relative expression of the relevant gene in the two sources. A relative optical signal, generated by the label in the duplexes, is determined. Independent claims are also included for the following: (1) similar method to M1 for isolating (I) derived from differentially expressed genes; (2) similar method to M1 for determining relative abundance of gene products; (3) similar method to M1 for isolating (I) according to the abundance of the genes from which they are derived; (4) composition comprising a mixture of microparticles that carry many identical single-stranded nucleic acids, different for each particle, and consisting of an oligonucleotide tag attached to (I) derived from a mRNA of at least one cell or tissue source; (5) kit for producing (B) consisting of microparticles each carrying a different oligonucleotide tag complement; and (6) kit for analyzing differential expression consisting of microparticles as defined in (4).
