Methods of producing novel enzymes

There is a described a method for producing modified enzymes which comprises modifying at least one residue in either the scaffold or active site lid of the a … barrel. In particular, the modification takes place in either the N-terminal segment, …1- a 1 loop, …6- a 6 loop or C-terminal segment. Typically the modification is achieved by targetted mutagenesis, recombination and selection. Modification of the bifunctional enzyme phosphoribosyl anthranilate isomerase (PRAI) and indoleglycerol phosphate synthase (IGPS) in E.coli and the grafting of a PRAI active site lid onto IGPS scaffold is described.
