Guanosine - 5<SP>1</SP> - monophosphate is prepared by reacting a 2<SP>1</SP>,3<SP>1</SP>-o-alkylidene guanosine with from 1 to 2 molar equivalents of tetrachloropyrophosphate per mole of guanosine in the presence of phosphorus oxychloride. The amount of tetrachloropyrophosphate used is suitably from 1 to 1À25 molar equivalents per mole of guanosine, and the alkylidene group may be isopropylidene,2-butylidene or 3-pentylidene. The 5<SP>1</SP>-guanylic acid is preferably isolated as the guanylic acid dioxanate, which may-be decomposed by heat to yield the free acid. In another method, the dioxanate is dissolved in water and one equivalent of a metal base, suitably an alkali or alkaline earth metal base, e.g. Na 2 CO 3 , KOH, CaCO 3 and Mg(HC0 3 ) 2 , is added and the water/dioxane mixture is then removed by evaporation under reduced pressure to yield the corresponding acid salt. |
