PURPOSE: Provided is a mutant strain SM5 (KFCC-11112) of Corynebacterium glutamicum (KFCC 10656) which is resistant to mercaptopurine in order to generate glutamic acid in a high yield. CONSTITUTION: A mutant SM5 (KFCC-11112) of Corynebacterium glutamicum is obtained by the following steps of: i) irradiating ultraviolet to Corynebacterium glutamicum (KFCC 10656) and treating bacteria with N-methyl-N'-nitro-N-nitrosoguanidine (NTG) to trigger mutation; and ii) selecting strains which can survive on a medium containing 10g/L of glucose, 1g/L of monopotassium phosphate, 0.5g/L of dipotassium phosphate, 0.5 g/L of ammonium sulphate, 1g/L of urea, 20m/L of iron sulphate, 0.5g/L of magnesium sulphate, 20m/L of manganese sulfate, 200μg/L of biotin, 5g/L of treacle, and 50-1000mg/L of mercaptopurine at pH 7.
