RECOMBINANT FUNGUS STRAIN FOR EXPRESSING BOVINE LACTOFERRIN AND BOVINE LACTOFERRIN OBTAINED THEREFORM

PURPOSE: Provided is a recombinant fungus strain which generates lactoferrin having excellent antibacterial activity in a high yield so as to be used in manufacturing medicines and protecting livestock from various diseases. CONSTITUTION: The recombinant fungus strain SKM2 (KCTC 0682BP) is obtained by the next steps of: i) selecting and cultivating strain RIB40 unable to generate arginine dehydrogenase; ii) obtaining RNA from bovine tissues and multiplying RNA through RT-PCR to separate cDNA; iii) multiplying only cDNA segments through PCR, which expresses lactoferrin; iv) ligating lactoferrin genes with vector pFGC33 and transforming strain DH5-α with the vector; v) separating recombinant vector pMKLF from the transformants; vi) making selection marker pDE·argB; and vii) transforming strain RIB40 with the recombinant vector pMKLF and the selection marker to obtain strain SKM2. The recombinant bovine lactoferrin is mass produced by fermentation culturing SKM2 in DPYA(dextrin polypeptone yeast extract5 ammonium sulfate) medium containing PMSF(phenylmethylsulfonyl fluoride) and EDTA(ethylenediaminetetraacetic acid).
