PURPOSE: To obtain a new erythropoietin compound having high in vivo bioactivity and useful for the therapy and/or prophylaxis of anemia, or the like, of chronic renal failure patients. CONSTITUTION: This new erythropoietin compound is a complex containing an erythropoietin glyco-protein having at least one free amino group, having an in vivo bioactivity increasing the production of reticullocyte, or the like, in myeloid cells and selected from human erythropoietin, or the like. The above glyco-protein is combined through a covalent bond with an 'n' polyethylene glycol group of the formula CO-(CH2)x-(OCH2CH2)m-OR (R is a lower alkyl; X is 2 or 3; (m) is about 450-900 and (n) is 1 to 3 with the proviso that (m) and (n) are selected in such a way that a difference in mol.wt. between the complex and the erythropoietin glyco-protein is 20-100 kDa). CO of each polyethylene glycol group is combined with one of amino groups mentioned above to form an amide linkage in the complex. The new compound is obtained by the condensation reaction of a compound of the formula with the erythropoietin glyco-protein. |
