PURPOSE: Disclosed is a simultaneous separation method of macromolecules and micromolecules using an on-line reciprocating size exclusion chromatography. CONSTITUTION: The equipment system for separation is composed of a tank (10), a pump (31,32), a column (40) of size exclusion chromatography, a valve (21,22), a solvent tank (51), a tank (52) for macromolecule, a tank (53) for micromolecule, and a timer. Using the chromatography, the operation steps are carried out continuously by the following procedures: a solvent recovery step (a fair direction), a macromolecule recovery step (a fair direction), a mixture recovery step (an inverse direction), and a micromolecule recovery step (an inverse direction). The packing material for the column (40) is one of silica, Dextran, Sephadex, or Bio-Gel P.
