The invention describes a method for isolating, from a naïve polypeptide repertoire which has not been preselected with a specific target ligand, a polypeptide of interest capable of interacting with said specific target ligand, which method comprises direct screening of the naïve polypeptide repertoire with the target ligand in order to identify the polypeptide of interest.
