Method, which does not require a culture stage, for predicting resistance to isoniazide in mycobacteria, isolated from clinical material, based on detecting a mutation in codon 315 of the katG resistance gene. A part of this gene is amplified by nested polymerase chain reaction (PCR) and the mutation is detected using a restriction endonuclease (RE). The outer pair of primers is CGAACCCGAGGCTGCTCC CACCCGCAGCGAGAGGTC and the inner pair TGGAGCAGATGGGCTTGG CAGTGGCCAGCATCGTCG. An Independent claim is also included for a reagent kit for the process containing at least the primers and/or RE. |
