The present invention relates to methods of inducing immunological tolerance in an animal receiving xenogeneic cells and tissues, especially by way of transplant, said methods comprising myeloablative or non-myeloablative conditioning regimens and administration to the recipient of a therapeutically effective amount of a mixture of transforming growth factor- beta (TGF- beta ) antagonists and tumor necrosis factor- alpha (TNF- alpha ) antagonists. The methods also include the use of anti-T cell antibodies as well as co-stimulatory blockade molecules such as anti-CD40 Ligand antibody and CTLA4lg.
