发明名称 NOVEL STAPHYLOCOCCUS HAEMOLYTICUS L62 (KCTC 8957P) GENERATING LIPASE
摘要 PURPOSE: Provided is novel Staphylococcus haemolyticus L62 (KCTC 8957P) generating lipase whose enzyme activity is increased 5-6 times by nonionic surfactant, especially at low temperatures, and which is useful in producing unsaturated fatty acids or synthesizing esters and peptides. CONSTITUTION: Novel Staphylococcus haemolyticus L62 (KCTC 8957P) and lipase generated thereby are obtained by the next steps of: i) selecting bacteria able to generate lipase from soil sample and identifying bacteria showing good enzyme activity from supernatant obtained after broth cultivation; ii) cutting the bacteria, S. haemolyticus L62, with restriction enzymes Clal and EcoRV, ligating DNA segments with plasmid pBluescriptII SK cut by the same restriction enzymes; iii) transforming E. coli XL Blue with the plasmid and obtaining five colonies showing lipase activity; and iv) separating plasmid DNA from the colonies and obtaining recombinant plasmid pSHL having 4.2 kb foreign DNA. L62 lipase has preprolipase consisting of signal sequence of 60 amino acids, pro peptide of 259 amino acids, and mature lipase of amino acids.
