PURPOSE: A method for producing a recombinant protein specifically binding with a human glucagon receptor and the use thereof are provided, thereby the human glucagon receptor can be easily separated without losing its activity. CONSTITUTION: The method for producing the recombinant protein comprises the steps of: fixing the human glucagon receptor or its mutated proteins as affinity ligands on a stationary phase; adding testing samples into a mobile phase contacted with the stationary phase; and obtaining the desired protein being capable of specifically binding with the human glucagon receptor, in which the affinity ligands have the amino acid sequence of sequence ID No. 2, and the stationary phase is a resin within chromatography. The expression vector pGR120HIS contains the gene encoding the amino acid sequence of sequence ID No. 2. E. coli (KCTC 0583BP) is a transformant transformed with pGR120HIS. The glucagon receptor fragment is produced by incubating E. coli KCTC 0583BP. |
