PURPOSE:To obtain a new sorbitol dehydrogenase produced by the culture of microbial cell belonging to the genus Pseudomonas and capable of producing sorbitol dehydrogenase, extremely useful for the enzymatic assay of D-sorbitol and usable e.g. for the diagnosis of diabetes. CONSTITUTION:This new sorbitol dehydrogenase useful e.g. for the enzymatic assay of D-sorbitol is produced by culturing a microbial strain belonging to the genus Pseudomonas and capable of producing sorbitol dehydrogenase [e.g. Pseudomonas sp.KS-E1806 (FERM P-14229)] in a medium. The enzyme is effective to oxidize D-sorbitol in the presence o NAD to produce D-fructose and NADH and specifically act on D-sorbitol and galactitol, has an optimum pH of about 10 and stable pH of 5.5-10.5 (treatment at 30 deg.C for 4hr), completely deactivated at pH <=5.5 and pH >=11.0, has optimum working temperature range of about 50 deg.C in 100mM tris-HCl buffer solution (pH 9.0), is strongly inhibited with HgCl2 and has a molecular weight of about 64,500+ or -5,000 (gel- filtration). |
