A method of genotype analysis in sheep to determine the PrP allele at one or more of amino acids 171, 154 or 136, said method comprising the following steps: a) obtaining a sample of genomic DNA or a copy thereof from a test animal; b) providing pre-determined primers to the target DNA sequence which vary in the final two nucleotides, the penultimate nucleotide being located at the polymorphic site and incubating with said sample under conditions suitable to allow polynucleotide binding; c) subjecting said incubated sample to PCR; d) determining the presence or absence of amplified DNA.
