摘要 PROBLEM TO BE SOLVED: To obtain the subject new enzyme derived from HCV polymerase N5B, having HCV polymerase activity, consisting of a polypeptide having a specific amino acid sequence, suitable for crystal structural analysis, and capable of e.g. designing HCV polymerase inhibitors. SOLUTION: This new enzyme is such as to be derived from HCV polymerase NS5B, have HCV polymerase activity, consisting of a new polypeptide having an amino acid sequence of the formula: X-Y [X is a consecutive amino acid sequence as a part of NS5B; the amino acid on the N-terminal of X is that at 1-position (Ser) on the NS5B, and the amino acid residue on the C-terminal of X is any amino acid residue at 531-position (Lys) to 570-position (Arg) on the NS5B; Y is carboxy group or another amino acid sequence not derived from the NS5B; one or more amino acid(s) in the amino acid sequence of X may be mutated or substituted with methionine or selenomethionine]. This new enzyme is useful for designing and/or identifying HCV polymerase inhibitors, developing HCV inhibitors, etc.
