A method whereby a gene mutation, which is useful in diagnosing cancer, etc., can be conveniently and exactly detected and quantitated. More particularly, a novel method of quantitating a gene mutation whereby the carcinogenic risk of a trace chemical can be evaluated by using a gene mutation concerning various diseases such as cancer as an indication and a further minor gene mutation, compared with conventional methods, can be detected and quantitated. A test gene is hybridized with a mutation detecting primer with the deletion of the site corresponding to the point mutation detecting site of the test gene at the 3'-end thereof. Then a radioisotope-labeled dideoxynucleotide is complementarily added thereto in the presence of a thermophilic DNA polymerase and the radioisotope of the dideoxynucleotide added to the site corresponding to the above-described mutation detecting site is analyzed.
