发明名称 IN VIVO SCREENING METHODS FOR PREDICTING CALCIFICATION OF IMPLANTABLE PROSTHETIC MATERIAL
摘要
An in vivo screening method reliably predicts calcification of implantable prosthetic material by implanting the prosthetic material intramuscularly in an animal. Calcification of the material is much more rapid when implanted in the muscle, as opposed to subcutaneous implantation, because of the greater calcium flux within muscle tissue, so that after a specified implant duration, the implanted material has relatively high calcification. The higher calcification provides greater resolution in the results. For example, calcification of intramuscularly implanted material may be on the order of 200 ν µg Ca/mg of dry tissue after 90 days when implanted in a rabbit, as opposed to less than 10 ν µg Ca/mg of dry tissue when implanted subcutaneously in a rat. Prosthetic material such as bovine pericardium tissue is implanted in the paravertebral muscle of a small animal such as a rabbit. Incisions are made through the skin of the animal, and a pocket is then formed in the muscle through each incision. The prosthetic material under test is then positioned within the pockets, and the incisions are closed. After the specified implant duration, the material is explanted and analyzed.
