PROBLEM TO BE SOLVED: To provide a method for transducing mutation to a template double strand nucleic acid by cleaving the template double strand nucleic acid to random fragments followed by adding a nucleic acid containing the range homologous or nonhomologous to the template to generate a single strand nucleic acid, annealing and carrying out polymerase treatment. SOLUTION: This method is a method for transducing at least one of mutation to a template double strand polynucleotide. The template double strand polynucleotide is first cleaved to a prescribed size of double strand random fragments and at least one of single or double strand oligonucleotide is added to the resultant double strand fragment group. In this case, this oligonucleotide contains the range homologous or nonhomologous to the template polynucleotide. By degenerating the resultant mixture to single strand fragments, annealing the fragments in a homologous domain between the single strand fragments, incubating together with a polymerase under a condition of forming mutagenized double strand nucleic acids and repeating these operations, the mutation is transduced into the template double strand nucleic acid. |
