PROBLEM TO BE SOLVED: To determine the ratio of allele from a reaction pattern by amplifying a double-strand DNA containing a polymorphic gene locus in a specimen using a primer pair and reacting the amplified product with a probe immobilized on a solid carrier. SOLUTION: In the amplification of a double-strand DNA region containing a polymorphic gene locus in a specimen using the 1st and the 2nd primer pair, the 1st primer locus specifically amplifies the 1st allele form of the polymorphic gene locus, the 2nd primer pair specifically amplifies the 2nd allele form of the polymorphic gene locus, each primer pair contains upstream and downstream primers, the upstream primer of each primer pair is complementary to a DNA chain opposite to the DNA chain complementary to the downstream primer, an amplified product is formed by using a primer labeled with different colored tag, the product is hybridized to two or more probes immobilized to known positions on a solid carrier and the ratio of allele on the polymorphic gene locus is determined from the pattern.
