Heterocyclic compounds of formula 1 for modulating methyl modifying enzymes such as histone methyl modifying enzymes, compositions and uses thereof are provided herein. An example of the compounds is N-((4-methoxy-6-methyl-2-oxo-1,2-dihydropyridin-3-yl)methyl)-6-methyl-7-(1-((R)-morpholin-2-yl)ethyl)-7H-pyrrolo[2,3-d]pyrimidine-5-carboxamide.
