Coupling a peptide (I) to a nonpeptide compound (II) having a carboxy or hydroxy functional group comprising forming a hydrazide linkage between (I) and (II) in solution, is new. Independent claims are also included for the following: (1) a modified peptide comprising a peptide coupled to a carboxy- or hydroxy-functional compound through a hydrazide linkage; (2) a synthetic vaccine comprising at least one modified peptide as in (I); (3) a diagnostic reagent comprising at least one modified peptide as in (I); and (4) use of N,N',N'-tri(t-butoxycarbonyl)hydrazine-N-acetic acid for functionalizing a peptide with a hydrazinoacetyl group, either at the N terminus of the peptide or at the end of a lysine or ornithine side chain at any position in the peptide sequence.
