A method of, and apparatus for, detecting the presence of an analyte in a sample, the method comprising: a) contacting the sample with a binding reage nt having a binding specificity for the analyte and an analyte receptor located within a test zone to form a first complex comprising the binding reagent, analyte, and analyte receptor, the first complex providing a test signal; (b ) contacting the sample with a reference ligand to form a second complex comprising the binding reagent, which further has a binding specificity for a reference ligand, and the reference ligand, the second complex providing a reference signal, wherein the reference ligand is located within a reference zone; whereby the ratio of the test signal to the reference signal defines t he level of analyte in the sample, this level being independent of the quantity of binding reagent contacted with the sample.
