Nucleic acid (I) encoding a eukaryotic neutral cerebral sphingomyelinase (II; nSMase2) that contains specific amino acid motifs (A1 and A2). A1 and A2 have sequences X1-X2-X3-X4-Asp-Tyr-X5 (A1) and X6-X7-Thr-Asp-His-X8 (A2) where X1, X6 = Ala or Gly X2, X3 = Arg or Lys X4, X5, X7, X8 = Ile, Leu, Val, Met Independent claims are also included for: (a) nucleic acid (Ia) that is complementary to (I); (b) derivatives, fragments and variants of (I) and (Ia); (c) (II) produced by expression of (I) or its variants; (d) antibody (Ab) directed against (I) or (II); (e) cell lines that overexpress (II); (f) transgenic mammals which overexpress (II) or have a genetic defect or deficiency of (II); (g) pharmaceutical or diagnostic composition containing (II), (I) and/or Ab plus auxiliaries; (h) screening for agents that alter expression or activity of (II); (i) producing (II) by chemical synthesis or recombinant expression in genetically altered organisms; and (j) producing (I) by chemical synthesis or replication in genetically modified organisms.
