摘要 PROBLEM TO BE SOLVED: To provide a method for simply, rapidly and remarkably amplifying a specific RNA sequence originally present in a sample at nearly constant temperature. SOLUTION: Inosine triphosphate is added as a substrate for an RNA polymerase in addition to adenosine triphosphate, uridine triphosphate, cytidine triphosphate and guanosine triphosphate in an RNA amplifying step for using a specific RNA sequence in a sample as a template, producing a double-stranded DNA containing a sequence homologous with the specific RNA sequence and a sequence complementary thereto and having a promoter sequence capable of transcribing the sequence, producing an RNA transcript by an RNA polymerase and further providing the RNA transcript for the template of the double-stranded DNA in a specified nucleic acid analysis in the sample expected to contain a specific nucleic acid. Thereby, the efficiency of the amplifying reaction is improved.
