摘要 PROBLEM TO BE SOLVED: To provide a method for enhancing the stability of a serine protease by correlating a metal ion-binding site of a protease to a protease stability parameter, and by enhancing the electrostatic attraction to the metal ion by the mutation of the sequence. SOLUTION: This is a method for enhancing the stability of a protease other than subtilin BPN' mutants in (1) Ser218 and Asp131, (2) Ser218, Asp131 and Ala254, (3) Thr116 and Asp131, (4) Asp131, (5) Ala169, (6) Ser169 by correlating a metal ion-binding site of a serine protease containing a metal ion-binding site to a protease stability parameter, and by enhancing the electrostatic attraction between the amino acid residue and of the metal ion-binding site and a metal ion by changing the structure of the metal ion-binding site by substituting, inserting, and/or deleting one or more amino acid residue(s) existing at the metal ion-binding site and/or in the vicinity of the metal ion-binding site.
