Allergen activity in dust is obtained by extracting from a dust sample at least one breakdown component of proteins or peptides; reacting the extracted at least one breakdown component with typically a colorimetric amine detection reagent such as TNBSA; and quantitatively measuring the intensity of any resulting coloration. Allergen activity is gauged by the intensity of coloration. Further, claimed are kits for obtaining the activity. Also claimed is a method for determining the activity using a protease substrate with immobilised proteins or peptides thereon labelled with a chromogenic compound. Utility is in obtaining allergen activity of house dust mites.
