摘要 A polynucleotide fragment (N1), comprising a sequence encoding a polypeptide (P1) having heparanase activity, is new. Independent claims are also included for the following: (1) a single stranded polynucleotide fragment complementary to at least a portion of N1; (2) a vector comprising N1; (3) a host cell comprising an exogenous N1; (4) a host cell particularly an insect cell, expressing a recombinant heparanase; (5) a recombinant protein comprising P1; (6) a polynucleotide sequence having the 1721 (I) or 1899 (II) bp sequences fully defined in the specification or a sequence homologous to (I) or (II); (7) a polynucleotide fragment capable of hybridizing with nucleotides 1-721 of (I); (8) a polypeptide sequence having the 543 (III) and 592 (IV) amino acid sequences fully defined in the specification or a sequence homologous to (III) or (IV); (9) a heparanase overexpression system comprising a cell overexpressing heparanase catalytic activity; and (10) a method for identifying a chromosome region harboring a human heparanase gene in a chromosome spread, comprising: (a) hybridizing the spread with a tagged polynucleotide probe encoding heparanase; (b) removing excess probe; (c) searching for signals associated with the tag. ACTIVITY : Antiangiogenic; vasotropic; Antiinflammatory; Antiarteriosclerotic. MECHANISM OF ACTION : Catabolic heparanase cleaves heparan sulphate chains.
