NOVEL FIBRIN DISSOLVING ENZYME

PURPOSE: Provided is novel fibrinolase effectively decomposing fibrin and having little toxicity. Also, its purifying method from Korean Salmosa Snake and its use as a thrombosis therapeutic agent are provided. CONSTITUTION: The novel fibrinolase shows the following characteristics; i) its molecular weight is 16,000-17,000 Da when measured by SDS-polyacrylamide gel electrophoresis and 31,000-33,000 Da when measured by gel filtration chromatography; ii) it is an acid protein and its pH is 4.6; iii) its activity increases in the presence of 2mM CaCl2 and decreases in the presence of 10mM EDTA; iv) maximum activity is shown at pH 7.5 at 37 deg.C; v) it hydrolyzes D-Phe-Pipecolyl-Arg-pNA and its activity is inhibited by DFP; and vi) it has affinity with fibrinogen and fibrin. A method for purifying the fibrinolase is characterized by the next steps of: i) separating crude toxin from Korean salmosa snake and diluting the toxin in Tris·HCl buffer solution; and ii) separating the toxin through a series of processes including ion exchange and gel filtration chromatography.
