METHOD FOR PURIFYING HEPATITIS C VIRUS PROTEINASE, NS3

PURPOSE: A method for purifying proteinase NS3, which decomposes the non-structural protein of hepatitis C virus, is provided for mass-producing NS3 proteins required to identify secondary structure and tertiary structure thereof. Thereby, an inhibitor for proteinase NS3 can be developed. CONSTITUTION: The method for purifying hepatitis C virus proteinase NS3 comprises the steps of: passing supernatant containing the hepatitis C virus proteinase NS3 through the SP sepharose column to purify the proteinase firstly; loading the purified hepatitis C virus proteinase into gel filtration chromatography column to purify the proteinase secondly; and optionally purifying the proteinase by cationic exchange chromatography, in which a buffer solution used in the chromatography contains 20 to 80 mM(pH 6.5) of phosphate chloride, 5 to 20 v/v% of glycerol, and 1 to 10 mM of DTT.
