EXPRESSION OF HUMAN INTERLEUKIN-4 IN YEAST

PURPOSE: Provided is a method for expressing human interleukin-4 (IL-4) in yeast to manufacture human IL-4 in a high yield by transforming yeast with an expression vector. CONSTITUTION: The method for expressing human interleukin-4 (IL-4) in yeast comprises the following steps of: i) designing base sequences of human interleukin-4 gene; ii) making an oligo nucleotide consisting of 20 bases; iii) synthesizing the whole DNA of double strand IL-3 gene through polymerase chain reaction (PCR); iv) inserting synthesized IL-4 gene into vector M13 and sequencing the gene; v) making expression vector M13 rt2-IL-4; vi) cutting the expression vector with restriction enzymes to obtain segment IL-4-T2/L and transforming E. coli with the segment to obtain plasmid pYLBC-A/G-UB/IL-4; and vii) transforming yeast with recombinant plasmid pYLBC-A/G-UB/IL-4 and cultivating transformed yeast.
