BASE SEQUENCE OF INTERNAL TRANSCRIPTION REGION OF MYCOBACTERIUM FORTUITUM

PURPOSE: The base sequence of internal transcription region of Mycobacterium fortuitum is provided which is used in producing oligonucleotide probe and primer. The probe and primer are used in diagnosing and identifying the species of Mycobacteria. CONSTITUTION: The base sequences of internal transcription(ITS) region of Mycobacterium fortuitum is represented by the sequence number 1. The probes and primers used for identifying Mycobacteria will be designed based on the ITS base sequence of Mycobacterium fortuitum. The ITS base sequence is useful in distinguishing genotypes since it has more numerous polymorphism regions as well as conservative regions than 16S rRNA commonly used as a target DNA. The base sequences of M. fortuitum and M. chelonae are identified, and probes and primers are synthesized using their base sequences. The probes and primers for other Mycobacteria are designed by analyzing the base sequence obtained from public domain(Genbank) with multialignment and blast and by selecting distinguishable regions by polymorphism. Then the primers and probes are identified by PCR and hybridization.
